This important histochemical substrate is used to detect the β-galactosidase enzyme in a range of scientific applications. Upon hydrolysis, X-GAL yields a localized, insoluble blue precipitate making it exceptionally useful in blotting and immunocytochemical assays. It is used to distinguish recombinant plasmids from parental vectors in cloning experiments using vectors containing the lacZ or lacZ "-peptide gene.  X-GAL has also been used for the detection of total coliforms in potable water and food products. X-GAL is often used in conjunction with IPTG (Isopropyl-β-D-thiogalactopyranoside) which binds and inhibits the lac repressor.

APPLICATIONS

1. Efficient in the rapid detection of coliforms in water and food with minimal false positives or false negatives.
2. Useful for dist inguishing between recombinant bacteriophages and the parental vector.
3. Suitable for the detection of lacZ reporter gene and colony identification.
4. Effective for the determination of localized enzyme activity in tissue sections.
5. Non-isotopic detector of inserts in cloning for histochemical marker genes.
