Intended use This immunoassay kit allows for the in vitro quantitative determination of mouse CD*4 concentrations in cell Lysates and other biological fluids. Introduction CD*4 is a cluster of differentiation molecule present on certain cells within the human body. It is a cell surface glycoprotein and functions as a cell-cell adhesion factor. It may also mediate the attachment of stem cells to bone marrow extracellular matrix or directly to stromal ***** expressing CD*4 (CD*4+ cell) are normally found in the umbilical cord and bone marrow as hematopoeitic cells, endothelial progenitor cells, endothelial cells of blood vessels but not lymphatics (except pleural lymphatics), mast cells, a sub-population dendritic cells (which are factor XIIIa negative) in the interstitium and around the adnexa of dermis of skin, as well as cells in soft tissue tomors like DFSP, GIST, PNSTs etc. Test principle The microtiter plate provided in this kit has been pre-coated with an antibody specific to CD*4. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for CD*4. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain CD*4, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of **0 nm 2 nm. The concentration of CD*4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
