Intended use This immunoassay kit allows for the use in vitro quantitative determination of human estrogen, E concentrations in cell culture supernates, serum, plasma and other biological fluids. Introduction Natural estrogens are hormones secreted principally by the ovarian follicles and also by the adrenals, corpus luteum, placenta . Exogenous estrogens (natural or synthetic) elicit, to varying degrees, all the pharmacologic responses usually produced by endogenous estrogens. Estrogenic hormones are secreted at varying rates during the menstrual cycle throughout the period of ovarian activity. The ***** of the anterior pituitary regulate secretion of the ovarian hormones, estradiol and estrogen; hypothalamic control of pituitary ***** production is in turn regulated by plasma concentrations of the estrogens and estrogen. During pregnancy, the placenta becomes the main source of estrogens. At the menopause, ovarian secretion of estrogens declines at varying rates. Test principle The microtiter plate provided in this kit has been pre-coated with an antibody specific to estrogen. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for estrogen and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain estrogen, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of **0 nm ± 2 nm. The concentration of estrogen in the samples is then determined by comparing the O.D. of the samples to the standard curve.
