Intended use This immunoassay kit allows for the in vitro quantitative determination of mouse Troponin T, Tn-T concentrations in serum, plasma and other biological fluids. Introduction Troponin T is a part of the troponin complex. It binds to tropomyosin, interlocking them to form a troponin-tropomyosin complex. The tissue specific subtypes are: Slow skeletal troponin T1, TNNT1 (*9q*3.4, *****1) Cardiac troponin T2, TNNT2 (1q*2, *****5) Fast skeletal troponin T3, TNNT3 (*1p*5.5, *****2) Test principle The microtiter plate provided in this kit has been pre-coated with an antibody specific to Tn-T. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Tn-T and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Tn-T, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of **0 nm ± 2 nm. The concentration of Tn-T in the samples is then determined by comparing the O.D. of the samples to the standard curve.
